MOLECULAR CHARACTERIZATION OF THE PHYTOOPATHOGEN Moniliophthora roreri, USING ISSR MARKERS, IN NORTE DE SANTANDER, COLOMBIA.

The regions of the department of Norte de Santander, Colombia, cocoa growers have the most serious disease that is the moniliasis caused by the phytopathogenic fungus Moniliophthora roreri. Initially the protocol of disinfection was carried out in the diseased ear, then it is sown in PDA agar (potato dextrose), modified PDA agar (gentamicin and cocoa pulp) and malt extract agar, incubating them for 10 days at a temperature of 15 ° C, showing better growth in PDA agar. After DNA extraction was continued, the effectiveness of conserved samples of isolated monilia in the municipalities of El Zulia, Cúcuta, Tibú, Puerto Santander, Bucarasica, El Tarra, Teorama and Sardinata de Norte de Santander, was later confirmed. the presence of DNA by agarose gel electrophoresis, 24 samples were selected, then standardized the ISSR technique (repeated inter sequences); for which they obtained band patterns in most of the 27 samples with the first 823, 880, 890 and 891, while with primer 816, 874 and 885 they generated very few bands. With the use of ITS4 and ITS5 the PCR was performed, sequencing was carried out on 2 isolated samples of the fungus and yield: for the HFa007 fungus with an identity percentage of 97.08% for Diaporthe pseudomangiferaem and for the HFa009 fungus a percentage of 99.29% Diaporthe melonis. The statistical analysis was executed, where a dendogram based on the Dice coefficient was obtained, which showed the existing relationships for the 28 samples studied. The 6d-17 sample from Sardinata was grouped independently.
